Anaplasma species of veterinary importance in Japan OAK

Anaplasma species of the family Anaplasmataceae, order Rickettsiales are tick-borne organisms that can cause disease in animals and humans. In Japan, all recognized species of Anaplasma (except for Anaplasma ovis) and a potentially novel Anaplasma sp. closely related to Anaplasma phagocytophilum have been reported. Most of these detected tick-borne pathogens are believed to be lowly pathogenic in animals in Japan although the zoonotic A. phagocytophilum has recently been reported to cause clinical signs in a dog and in humans. This review documents the studies and reports about Anaplasma spp. in Japan. c Ybanez and Inokuma.

Clinical usefulness of antibodies against Babesia gibsoni detected by ELISA with recombinant P50 OAK

The clinical usefulness of antibodies against Babesia gibsoni detected by ELISA with recombinant P50 was examined in dogs in an area where B. gibsoni infection was endemic. Only 8 among 14 dogs with acute type B. gibsoni infection without a previous history of infection were positive. This high percentage of false-negative results is thought to be a weak point of ELISA as a diagnostic tool. However, 14 other anemic dogs with a confirmed history of B. gibsoni infection were all positive, thus confirming the higher sensitivity of ELISA in detecting a history of infection.

Detection of Anaplasma bovis DNA in the Peripheral Blood of Domestic Dogs in Japan OAK

The prevalence of Ehrlichia and Anaplasma in 1,427 dogs from 32 Japanese prefectures was evaluated by PCR and DNA nucleotide sequencing. PCR screening demonstrated that 18 dogs (1.3%) were positive for Anaplasmataceae. Sequence analysis revealed that 14 of the amplicons were most closely related to Wolbachia spp., symbionts of Dirofilaria immitis, whereas three were identified as Anaplasma bovis. The remaining amplicon could not be sequenced. Almost the entire sequence of 16S rRNA (1,452 bp) from one of the positive specimens was determined, and subsequent phylogenetic analysis confirmed that the detected sequence was that of A. bovis. Detection of Babesia canis rossi, B-canis vogeli, and Hepatozoon canis in dogs in a village of eastern Sudan by using a screening PCR and sequencing methodologies OAK

Babesia and Hepatozoon infections of dogs in a village of eastern Sudan were analyzed by using a single PCR and sequencing. Among 78 dogs, 5 were infected with Babesia canis rossi and 2 others were infected with B. canis vogeli. Thirty-three dogs were positive for Hepatozoon. Hepatozoon canis was detected by sequence analysis.

Detection of Borrelia garinii, Borrelia tanukii and Borrelia sp. closely related to Borrelia valaisiana in Ixodes ticks removed from dogs and cats in Japan. OAK

Ticks removed from 1136 dogs and 134 cats all over Japan were examined for Borrelia infection by PCR and sequencing. The 5S-23S rDNA intergenic spacer of Borrelia was detected from two Ixodes persulcatus ticks from two dogs and two unidentified Ixodes spp. from another two dogs in Hokkaido, and two Ixodes granulatus ticks from two cats in Okinawa. Additional 2 I. granulatus from the same cats also showed positive. Detection of DNA of 'Candidatus Mycoplasma haemominutum' and Spiroplasma sp. in unfed ticks collected from vegetation in Japan OAK

DNA fragments of 'Candidatus Mycoplasma haemominutum', a feline heamobartonella pathogen, were detected from unfed Ixodes ovatus collected from vegetation in Hokkaido, Fukushima and Yamaguchi Prefectures, and unfed Haemaphysalis flava in Yamaguchi Prefecture. This finding suggests that ixodid tick is a possible vector of 'C. Mycoplasma haemominutum'. Spiroplasma DNA was also detected from unfed I. ovatus in Hokkaido, Fukushima and Yamaguchi Prefectures. The analysis of nucleotides sequence suggested that this Spiroplasma was distinct from registered species.

Detection of Rickettsial DNA in ixodid ticks recovered from dogs and cats in Japan OAK

DNA from ticks recovered from 1137 dogs and 133 cats from Ell over Japan were examined for Rickettsia infection by citrate synthase gene (gltA)-based PCR and partial nucleotide sequencing. A total of 91 dog tick samples and 18 cat tick samples showed a single band of the appropriate size in the nested PCR. Sequence analysis was successfully performed on 102 samples. DNA of Rickettsia japonica or closely related Rickettsia spp. strains were detected from 38 licks in 16 prefectures mainly in western Japan. To determine the distribution of Babesia gibsoni infection in dogs in the eastern part of Japan, an epidemiological survey of dogs suspected of having B. gibsoni infection was attempted using the polymerase chain reaction (PCR) and enzyme-linked immunosorbent assay (ELISA). Thirty-five of 115 such dogs (30.4%) were positive by PCR and/or ELISA. The 35 positive dogs consisted of 28 Tosa dogs, 4 American Pit Bull Terriers, and 3 mongrel dogs in Aomori, Fukushima, Ibaraki, Gunma, Chiba, Tokyo, Kanagawa, and Nagano Prefectures. Experimental inoculation of beagle dogs with Ehrlichia species detected from Ixodes ovatus OAK

Three beagle dogs were inoculated with mice spleen/liver homogenate infected with Ehrlichia species detected from Ixodes ovatus (EIO) and one dog was used as a control. All three infected dogs did not show clinical signs of disease except for mild pyrexia throughout the 41-day study period. Splenomegaly was observed from Day 7 post-inoculation (p.i.) in two of the dogs. Hematological and biochemical abnormalities included mild thrombocytopenia, hypoproteinaemia, hypoalbuminaemia and increased C-reactive protein values. High Genetic Diversity of Anaplasma marginale Detected from Philippine Cattle OAK

A total of 658 cattle in 6 provinces in the Philippines were screened for Anaplasma marginale infection by using a diagnostic heat-shock operon (groEL) gene-PCR assay. The screening-positive samples were further tested using the major surface antigen protein 1a (Msp1a) gene-PCR assay. Screening PCR results showed 130 cattle (19.8%) were positive for the A. marginale infection. Subsequent amplification using the Msp1a gene only showed 93 samples (14.1%) to be positive. Molecular Survey of Rickettsial Agents in Feral Raccoons (Procyon lotor) in Hokkaido, Japan OAK

Rickettsial infection in feral raccoons (Procyon lotor) in Hokkaido, Japan was analyzed by molecular methods. Genus-specific nested polymerase chain reaction (PCR) analysis based on the Rickettsia citrate synthase (gltA) gene showed that 13 of 699 raccoons (1.9%) examined were positive for Rickettsia. Twelve of the 13 partial gltA sequence amplicons were successfully analyzed. The nucleotide sequence of one amplicon was identical to both Rickettsia heilongjiangensis and R. japonica, one was identical to R. felis, and the rest to R. helvetica. This is the first report on the detection of rickettsial agents in peripheral blood of raccoons.

Multiple infections of Anaplasma platys variants in Philippine dogs OAK

Perforating abomasal ulcer caused by yolk sac tumor in a Holstein calf OAK

A 2-month-old male Holstein calf showed clinical signs of abdominal bloating, melena, and pain and was suspected of having a perforating abomasal ulcer. Necropsy revealed a large mass located preferentially around the abomasum and a large perforating abomasal ulcer on the pyloric antrum. Milky white masses of various sizes were also found in the abdominal cavity that consisted of agglutinated nodules ranging in size from a pinhead to a golf ball and were distributed on the surfaces of the liver, reticulum, omasum, abomasum, and diaphragm. Pericardial mesothelioma with severe congestive heart failure in a Holstein cow OAK

A 58-month-old Holstein cow showed anorexia, edema of the lower jaw and dewlap, jugular venous engorgement, abdominal gas, and watery diarrhea. Heart sounds were faint on auscultation, and decreased rumen motility was noted in physical examination. Echocardiography findings included adhesion of fibrin-like structures to the pericardium, highly echogenic periaortic region, and pericardial effusion, which suggested traumatic pericarditis or tumor formation. Although atypical mesothelial cells were observed in bloody pericardial fluid, no diagnosis was made. At complete necropsy, milky mass formation was observed on the epicardium. Polysplenia syndrome with duodenal and pancreatic dysplasia in a Holstein calf : A case report OAK

Background: Laterality disorders of the abdominal organs include situs inversus totalis that mirrors the arrangements of all internal organs and heterotaxy syndrome (situs ambiguus) in which the thoracic or abdominal organs are abnormally arranged. Heterotaxy is often accompanied by multiple congenital malformations, and it generally comprises asplenia and polysplenia syndromes. Retrospective study of clinical and laboratory findings of autosomal recessive cholesterol deficiency in Holstein calves in Japan OAK

This retrospective study was conducted to clarify the clinical findings of an autosomal recessive cholesterol deficiency disorder in Holstein calves. Thirty-three calves with poor growth by uncertain causes from 2007 to 2016 comprised the study population of calves that were homozygous (n = 12) and non-homozygous (n = 21) for the disease. No significant group-dependent differences were found in the proportion with diarrhea or fever. Fewer calves with ataxia and anorexia were found in homozygotes. All affected calves showed muscular atrophy. Median RBC, Hb, and Ht values in affected calves were significantly lower. Serological evidence of infection of Anaplasma and Ehrlichia in domestic animals in Xinjiang Uygur Autonomous Region area, China OAK

Serological methods were utilized to detect Anaplasma and Ehrlichia infection in domestic animals in Xinjiang Uygur Autonomous Region, China. By using an indirect immunofluorescence assay (IFA), antibodies that reacted with Anaplasma phagocytophilum and Ehrlichia chaffeensis were detected mainly in ruminants kept on pastureland in Altai, Ili and Kashgar area. Antibody titers up to 1:320 were recorded. These results indicate that ruminants kept in these areas may be infected with some species of Anaplasma and Ehrlichia.
